Optimization of Submerged Fermentation Parameters for Enhanced Lipase Production from Newly Isolated Lipolytic Yeasts using Agro Waste Substrates

Fungi are currently recognized as the most preferred sources of lipase due to their ability produce copious amount extracellular with lower production’s and extraction’s costs. Lipase production was carried out in agro wastes formulated-liquid fermentation media at 300C, 130rpm, initial pH 7 for 96 hours using newly isolated LipA gene identified yeasts [Pichia kudriavzevii ENS1 [Accession No: OL546800], Candida orthopsilosis PMS3 OL546803] Saccharomyces cerevisiae PMS8 OL546805] inoculum size 2%. Agrowastes such Mango kernel orange peel were used alternative carbon sources; beans hull groundnut shell nitrogen while lipidic olive oil, soyabeans palm oil coconut inducers. Single factor optimization process determine effects sources, inducers, incubation time, size, carbon: ratio on cell growth. Proximate analysis agro-waste substrates showed higher percentage carbohydrate crude fat mango powder 68.05±0.56% 19.54±0.62% recorded protein 11.63±0.05%. study optimum 50.83±1.18u/ml/min 0.69±0.00[OD] PMS3[Accession hot water treated hydrolysate source. The results from this that would be an economical source industrial-scale production.
